Abstract A highyielding enantioselective synthesis of the bioactive S—N5chlorothiophene2sulfonyl ββdiethylalaniol 7.b.2 a Notch1sparing γsecretase inhibitor metabolite with EC5028 nM effective in reduction of Aβ production in vivo has been realized starting from readily available 3pentanone. The key steps of the synthesis are prolinecatalyzed αaminooxylation and αamination of aldehyde the latter contributing an overall yield of 5075 and 9099 enantiomeric excess.
